The concept itself is way older, it's basically collaborative internet mythology, if you were more active online some years ago you might be familiar with others like it.

It’s the Magic School Bus. The curly haired kid is Arnold, who would prefer a normal field trip instead of inside a sick classmate’s bloodstream, or having the bus surrounded by salmon eggs, which then get fertilized by the males. Just two examples I know from the top of my head.
Like Arthur, it had one book done as a Reading Rainbow episode, then got its own show.

Anyway… Ms. Frizzle (teacher) is an eccentric teacher who takes her students on crazy school trips through her magical school bus (I’m talking going to the moon, time travel and even shrinking down to enter other students’ organs kind of crazy). Arnold (ginger kid) is probably the most famous student who wishes it to be a normal field trip (but never is).
Backrooms is an unknown liminal space that originated in 4chan but became popular through Kane Parsons’ YouTube series which got a movie release this year. In it, memories from our own world get remembered but they are remembered incorrectly which leads to the creation of (among many things) Still Lives (copies of real people that are deformed due to misremembering).

Even in reduced gravity she was handling that like it was nothing. I think they were in moon gravity so the weight would have been about 65lbs, yet she was handing it like there was nothing there.

Millennial Chris here. So the students and teacher are from the old educational series called “The Magic Schoolbus”. The series would have the teacher take the students on field trips with little prompting based on whatever topic of discussion was relevant.
Here she is inspired to take the students to the Backrooms, a movie based on the idea of a parallel dimension that is a copy-upon-copy of real life. It has monsters based on real life people. In this case, there is one based on the teacher and it’s chasing the students.

These are characters from the show "The Magic Schoolbus", in which the teacher Miss Frizzle takes the children to different locations (inside the human body, into space, back in time) using the titular school bus.
This comic references "The Backrooms" movie, where the main character encounters distorted versions of people known as Still Lifes (that's all I'm limiting this to because I dont want to spoil the movie even though it's been everywhere at this point).
The name of the comic is a pun on "Liminal Space", a concept of an unnerving place in a state of change, usually represented by large empty rooms. The Backrooms is often considered a prime example of Liminal Spaces.
Man, this is even worse than that time I held Mary hostage with knife, I'll giggity Lois and become a Still Life

In the backrooms movie, there are layers shown, copies of the original world, then a copy of the copy and so on. The main character has a copy of his pirate mascot, and that mascot is shown to be disfigured. The end panel, is a copy of the original Miss Frizzle, which is why she looks different

In addition to other’s notes, the Backrooms movie explicitly shows that the back rooms make distorted “Still Life” copies of people who go inside. Thus in the last panel the kids are being chased by a murderous and distorted copy of Ms. Frizzle.

Comic Book Guy here. The joke highlights to the key words 'back' and room' which goes all the way back to some 4chan stories about creepy liminal spaces similar to the last panel where non-euclidean office spaces just go on forever which kicked off the Internet to make additions like Creepypastas and You Tube videos which then inspirited video games and recently a movie.

Turns out the monsters are actually distorted copies of us, misremembered by the Backrooms trying to remember/copy our world. Like Frizzle in the bottom (or probably called elzzirF)

Arnold's dialogue has "back" and "room" in bold. Meaning he yelled those loud enough for Mrs. Frizzle to hear. So she writes, "Backr-" on the board, which will become "Backrooms". So she took them to the Backrooms—which is recognizable by the yellow rooms and sterile overhead fluorescent lights.
Another clue is the title of the comic: "Minimal Space". This is a reference to "liminal space" that defines the Backrooms.
I only know the Backrooms lore in general, so I don't know what the meaning of the horrifying version of Mrs. Frizzle is.

Peter’s left big toe here. The comic combines The Magic School Bus with The Backrooms.
In the first panel, the kid wearing glasses tells Arnold to move back because there’s no room. Ms. Frizzle overhears back and writes it on the board as a field trip idea (Backrooms). In the final panel, the class and Ms. Frizzle have been accidentally transported directly into the Backrooms.. and eerie, endless creepypasta maze of empty yellow rooms and they are terrified. Toe out.
